Retatrutide is a novel triple receptor agonist targeting GLP-1, GIP and glucagon receptors. In research it has demonstrated some of the largest weight-reduction effects reported for any incretin-class molecule, alongside improvements in glycemic control and hepatic fat.

Benefits

  • Pronounced reductions in body weight in trial subjects
  • Improved insulin sensitivity and fasting glucose
  • Reduced hepatic steatosis markers
  • Appetite suppression and slower gastric emptying

Considerations

  • GI side effects: nausea, vomiting, constipation, diarrhea
  • Possible elevated heart rate
  • Rapid loss of lean mass if calories/protein too low
  • Long half-life — side effects can linger

Protocol

Subcutaneous injection once weekly. Titrate slowly over 4–8 weeks to limit GI side effects. Cycle 12–24 weeks then assess.

Dosage

Typical research titration: 2 mg/week → 4 mg → 8 mg → 12 mg. Maximum doses studied up to 12 mg weekly.

Reconstitution

10 mg vial: add 2 mL bacteriostatic water (5 mg/mL → 0.4 mL = 2 mg). 15 mg vial: add 1.5 mL BAC water (10 mg/mL).

Always use bacteriostatic water (BAC), sterile technique and store reconstituted vials refrigerated unless otherwise noted.
